A range of full-length and partial CDNAs have been cloned from high altitude plant Rehum emodii and Caragana jubata 

 

Rehum

RACE yielded a bHLH fragment of 1.2 kb and 3’ RACE yielded a bHLH fragment of 0.8 kb. Using the sequence data, full-length cDNA was cloned (Fig.4.9). Sequence analysis of full-length gene showed it belonging to MYC-related DNA binding protein, known to function as transcriptional activator under dehydration and cold.

 

Further, full-length cDNAs of a MAPKinase were cloned. The kinase was found to respond to the temperature, ABA, jasmonate, salicylate and drought stress (Fig. 4.10).   Through differential display of mRNA, certain interesting genes such as for calmodulin, Mn-SOD, and Cu-ZnSOD were obtained. Using RACE, full-length cDNAs for these genes was obtained and cloned.

Cloning of Lipoxygenase fron Caragana

 

Partial fragment of LOXobtained after differential display was used to design primers for 5’ and 3’ RACE and finally a full-length cDNA was cloned (Fig. 4.11).
