Genetic diversity analysis of germplasm

Leaf samples from 13 trees growing across the country were analyzed using AFLP markers. A total of 174 polymorphic bands were obtained from four AFLP primer combinations and the relationship suggested a common source of introduction. AFLP and microsatellite markers were also used to depict the genetic relationship. These were similar in both the analyses.

Screening of ginkgolides and bilobalide

Chemical characterization of flavones and flavone glycosides Leaf samples were processed for characterization of ginkgolides, bilobalide and flavonol glycosides of kaempferol, quercetin and isorhamnetin. The contents of bilobalide, ginkgolide A, ginkgolide B, ginkgolide C and ginkgolide J estimated by HPLC-ELSD in the different samples showed variations in the content of ginkgolides and bilobalide. Variability of ginkgolides and bilobalide was also assessed in leaf samples collected from different locations.

A simple, sensitive and rapid HPLC-ELSD method was developed and validated for the quantitative determination of terpene trilactones (TTLs) and flavonoids. Of the different methods used, ultrasonic assisted extraction was the most efficient for the extraction of TTLs. The samples collected from 1,450-1,800m altitudes had higher TTLs and flavonoids
